I am by no means an expert on downtown stuff

If you want some upscale Canadiana: Canoe
Upscale mainstream Japanese: JaBistro
If you're meatetarian and would like an Oktoberfesty experience: WVRST
Pan-Asian fusion fastish food: Bahn-Mi Boys

If you can make a day-trip out to the east end, the Distillery District and St. Lawrence Market are pretty close together and have all sorts of stuff to try.
Might be worth getting a transit day-pass for such a trip and they can be snagged at Pearson or at subway stops once you're situated.
 
I second herman's love for Banh Mi Boys, as it's very tasty!

I'm most well versed in restaurants in Greektown, being a Greek myself :P, so if you make it out there, Mezes is my favourite.  Messini is great for Gyros as well.

Other than that, I'm very into the dive bar.  Sneaky Dee's is great if you want a plate of nachos and to check out a punk rock show, and is right around the corner from Kensington Market.  Defina in the Junction makes great pizza, and Barque is Roncesvalles is a terrific BBQ place.  The Lakeview Diner at Dundas/Ossington is a 24 hour diner, and great for brunch or lunch/dinner, and on the opposite corner is the Haifa Room, which is a Middle Eastern place that just opened - I was there last Thursday, and it was delicious!
 
I know you're only asking about food, but in case it appeals to your touristy needs, and given you're near the CN Tower:

Aquarium is very good and literally adjacent to CN Tower. CN Tower itself is fine but really only an hour's investment. ROM is good, Casa Loma maybe not worth the time. Science Centre is for all ages and great, but honestly not worth it without kids. Zoo is solid but a ways north of the city core and there's a lot of outdoor walking, best with kids of course.

I second Defina and Barque - though, they're not in the area you're staying, they're both walking distance from subway stations/streetcar lines.

If you make it to the Distillery District, I strongly recommend grabbing some hot chocolate from Soma.
 
It mostly depends on the trip purpose: business or family trip? A family trip will require some venturing out further to either west or east end. A lot of authentic immigrant-run little restaurants outside the downtown core (Danforth Ave). Some legit Scottish and Mexican and Somolian and Vietnamese and Filipino and Thai and Chinese and Indian and West Indies fare in Scarborough. Lots of similar ones in Etobicoke as well. Yonge street from Sheppard to well north of Steeles has all sort sorts of east Asian options (primarily Korean).

If you're on business and just want to juice the Canadian economy with some USD with the lads, check out Jacobs and Co. Steakhouse.
or the Copacabana for Brazilian barbecue.

I'll defer to others on pubs and night-time festivities.

I'm not Toronto based and like herman says there's probably a lot of "smaller" gems that you can find in the area but I have been to the Copacabana in Niagara Falls and holy hell was it some great food/a fun experience.

This, too. Toronto is bursting will all sorts of authentic international cuisine.
 
Of the neighbourhoods mentioned I'm most familiar with Kensington but I'd point out that while the market itself is a great spot for food, it's not necessarily a great spot for restaurants(as in, lots of good takeout and counters but not much in the way of sitdown ambiance). It's also not a spot I'd take to impress any business associates. That said in the general neighbourhood I'd recommend:

Rol San for all-purpose Chinese and Dim Sum
Pho Hung for Vietnamese
7 Lives for Tacos
Rasta Pasta for an interesting take on Jamaican
Egg Bae for Breakfast sandwiches
Rudy's for Burgers
Bar Raval for Tapas
Fresca Pizza for Pizza/Italian Sandwiches(Although California Sandiches, a little further west, is better for the sort of giant Veal/Chicken/Meatball Italian sandwiches that are a Toronto staple)

If you're looking for post-bar food many of the Chinese spots on Spadina are open very late and Pizza Gigi, a few blocks north of Kensington, is a solid option and delivers until 4 am.

My favourite two Thai restaurants in the city are right next to each other: Pai and Khao San Road. I really like Town Crier for pub/beer selection. Figo is pretty good for fancied up Italian, Rozina is really good for more Rustic Italian. Favourite Gelato in the city is Mizzica right off of Queen. For a more adventurous and fancy restaurant I liked Woods Restaurant.

Agreed that personally for me one of my favourite grab n go sandwiches is Banh Mi Boys.
 
Even worse news? I have officially bought TMLFans.ca. Starting tomorrow it is now exclusively limited to discussions about labour practices in sports and our favourite Wes Anderson movie soundtracks.
